In the Chinese Pharmacopoeia (2025 Edition), the quantification standard for Phyllanthi Fructus has been revised from calculation based solely on gallic acid to the total content of gallic acid plus ellagic acid. Meanwhile, the chromatographic elution mode is updated from isocratic elution to gradient elution, imposing stricter requirements on instrument stability. An HPLC method was established for the simultaneous determination of gallic acid and ellagic acid in Phyllanthi Fructus.
Analyses were performed with a C18 column following the chromatographic conditions specified in the 2025 Edition of Chinese Pharmacopoeia. In repeatability tests of reference standard solution, the RSD of retention time for gallic acid and ellagic acid was ≤0.187%, the RSD of peak area was ≤0.912%, and the theoretical plate number of gallic acid was 10622, all complying with pharmacopoeia specifications. This method can serve as a reference for routine content testing of gallic acid and ellagic acid in Phyllanthi Fructus.
