Protocol in Pharmacopoeia of the People’s Republic of China (2025 Edition) — Determination of Ginsenoside Rg1, Ginsenoside Rb1 and Notoginsenoside R1 in Notoginseng Radix et Rhizoma

The assay of Notoginseng Radix et Rhizoma is a revised item in the Pharmacopoeia of the People’s Republic of China (2025 Edition), in which the elution procedure is adjusted from a two-segment gradient to a four-segment gradient. In accordance with the chromatographic conditions specified in the 2025 Edition Chinese Pharmacopoeia, a method for the determination of ginsenoside Rg1, ginsenoside Rb1 and notoginsenoside R1 in Notoginseng Radix et Rhizoma was established using a C18 chromatographic column.

The results of repeatability tests on reference standard solution are as follows: The RSD of retention time for ginsenoside Rg1, ginsenoside Rb1 and notoginsenoside R1 ≤ 0.155%; The RSD of peak area ≤ 0.099%; Tailing factors range from 0.882 to 1.047; The theoretical plate number of notoginsenoside R1 reaches 7867, which meets the requirements of the Pharmacopoeia of the People’s Republic of China (2025 Edition). This method can provide a reference for the quantitative analysis of ginsenoside Rg1, ginsenoside Rb1 and notoginsenoside R1 in Notoginseng Radix et Rhizoma.
